Earlier this week, Biden spoke with Putin, a conversation the two leaders had by phone in three weeks as tensions rose in Ukraine.
Discussing a phone call with Putin on Friday, Biden made it clear he would not negotiate here.
"I'm not going to negotiate here in public, but we made it clear that he can't - I stress, can't - attack Ukraine," The Defense Post quoted him as saying.
The US leader also added that Washington and its allies would have tough sanctions if Russia dared to carry out an attack on Ukraine.
"Making it clear to President Putin that we will have severe sanctions, we will increase our presence in Europe, with NATO allies if Russia invades Ukraine," he said.
Not only that, the U.S. also provides serious support to build trust to reduce these tensions.
"Express support for confidence-building measures to reduce tensions in Donbas and active diplomacy to advance the implementation of the Minsk Agreements."
Washington and its European allies have recently accused Russia of threatening Ukraine with a new invasion.
About 100,000 Russian troops have gathered near the border between the two countries. Putin seized crimea in 2014.
Russia has also been accused of waging a pro-Russian separatist war that erupted that same year in the east.
